Ooo, I love moments of discovery! They're a reason to have a story...
I second the moment from Spider-Man 2 with MJ. That movie has a few. Harry finding out who Spider-Man is, the people on the train seeing Peter Parker, and the moment when Peter bursts up out of the pile of rubble, back in full strength after MJ was taken... he'd realized he needed to have a clear idea of what he really wanted and he'd just gotten it. Did I mention it's one of my favorite movies?
Some Kind of Wonderful - has a teaser and then a revelation. There's a moment when Watts almost gives herself away, and one when Keith sees everything crystal clear. But I won't give either away! If you've seen it, you know the best bits.
X-Men - Logan thinks Magneto has been chasing him? Haha, silly Logan, that's what happens when you give so many people reason... no, Magneto wanted sweet little Rogue.
The Shop Around the Corner - It's a spoiler... it includes this: "My dearest, sweetheart Klara, I can't stand it any longer. Take your key and open post office box 237 and take me out of my envelope and kiss me." Yeah, I know, weird. I'm a sucker for a GOOD romance. Not just any weak tired love story with the same old set-up so worn out that you don't even need to work at it anymore. A well-established bit of romance building up to that discovery. Yeah, that's the stuff.
MegaMind - I love this movie! There's a few in it... but again, for fear of spoilers, I'll be cryptic. Who's that bearded guy with the sandwich? And who is she, in fact, really kissing? And not to mention, who is REALLY rescuing Roxanne, and who is fighting Titan? Not to mention a couple of mushy ones...
A little more obscure... a couple from Bollywood:
Dilwale Dulhania Le Jayenge - When Raj finally stops joking and says, "Simran, I've fallen in love with you..." and then snorts and starts laughing and calls her a sucker. Only as it happens...
Rab Ne Bana Di Jodi - "Taani, partneeer!" Saying prayers the morning of the dance competition, then Taani's sorrowful (for her) speech right before it, followed by the dance sequence at the end. All very, very mushy stuff. Seriously, if you like a good love story and are cool with Indian musicals (and Shahrukh Khan), these last two will satisfy.
Fullmetal Alchemist Brotherhood - spoiler here, too, so watch it or read it first... When Ed realizes that Al is about to... well, whatever it was he did exactly in order to give Ed his arm back and save him... and is thrashing and screaming and cussing at him to stop when it happens, at which point he promptly channels all his rage into pounding the bad guy. Really a superior manga and anime.
And... "Yer a wizard, Harry." If I had a nickel for every time I've heard that... um, in the movie, I mean.
I could probably find more... I may later (yeah, that's right... it's my special interest! One of them.).
