Why should the gender matter?
I suppose it doesn't from the standpoint of the need to be nice. It's just that the social response to a male mascot character may be different from the response to its (likely) female occupant.
When I was a teenager we went to Chuck E Cheese's with my younger sister and at the time it was quite a shock for me to see the male character remove the head mask to reveal a female occupant. It's such a big contrast for how I would interact with the character.
