I think maybe because people with AS have a unique way of thinking.
In general terms, humour for NT's is when somebody says something unexpected or unique of makes an intelligent comment/observation about something that the NT may not have thought of.
So to an NT you are saying something unique, so it is seen as humour.
Age1600, when you asked your colleague why they smiled when talking on the phone ... well that is a good point. So in part, your colleague was laughing at herself.
I find my sons' very refreshing. They offer me an alternate viewpoint. They make observations that I would not make. Some of what they say makes me laugh, because they're pointing out something that I missed. When they point it out I think, 'Wow, that was so obvious ... aren't I a big silly!'
I think NT's are so busy 'following the crowd' they don't stop and think about what they're doing / why they're doing it. Observations by people with AS makes them stop and actually think.
I don't know if I'm explaining this well. I'm an NT Mum with 3 sons with AS.
My 3 sons (with AS) often say unexpected things (unexpected to an NT like myself) and I laugh. They get upset because they didn't mean to be funny.

I used to think my husband (who has AS) was mad a lot because his facial expression is so blank. I used to drive him crazy, asking him what was bothering him.
Now I will only ask him once if I think he's upset. If he says he's OK ... I leave him be.
