It's actually (and ironically) yet another cultural appropriation from the African American community.
It originates from a white woman who made up a lie about an African American boy by the Emmett Till whistled at her. The teenager was tortured and lynched. Although her name wasn't Karen, subsequently every white woman who made a false or exaggerated claim to the police about a black person was labelled "Karen".
In the last 10 years youtube videos of white women blatantly lying on camera have become viral sensations. The name is stereotypically white American but personally I prefer "Becky" or "Tammy".
